P450 cyptide synthase MpoB catalyzes the cross-linking of the YPW motif on the precursor peptide

The uncharacterized cytochrome P450 cyptide synthase MpoB from Micromonospora polyrhachis DSM 45886 catalyzes a cross-link between Tyr-C3 and Trp-N1 at the YPW motif on the precursor peptide MpoA.

Efficient synthesis of O-glycosylated amino acids

Synthetic glycopeptides are versatile tools to investigate protein glycosylation. In this work, we outline a short, economical synthesis to O-glycosylated amino acids compatible with Fmoc-SPPS.

Sources of mismeasurement of RNA knockdown by DNAzymes and XNAzymes

Nucleic acid catalysts can carry over into cellular RNA preparations following transfection, causing false positive and false negative knockdown measurements.

The impact of conjugation strategies and linker density on the performance of the Spermine-AcDex nanoparticle–splenocyte conjugate

A common approach in living medicine engineering is modifying cell surfaces with nanomedicines. The choice of conjugation strategy and the chemical structure of the linker are crucial for living medicine design.

Photoactivatable and photolabile pharmacophores: lessons learned from capsaicin

Structurally merging light-susceptible with bioactive molecules allows to generate photolabile chromopharmacophores. Using capsaicinoids, chromopharmacophores could activate the TRPV1 channel, while no activation is observed for the photoproducts.

Structure-guided engineering of a polyphosphate kinase 2 class III from an Erysipelotrichaceae bacterium to produce base-modified purine nucleotides

The polyphosphate kinase 2 class III from an Erysipelotrichaceae bacterium is highly promiscuous. We determined the first crystal structure of the enzyme and applied structure-guided engineering to enable efficient access to ATP and GTP analogues.

The SpyBLI cell-free pipeline for the rapid quantification of binding kinetics from crude samples

A less than 24-hour workflow with little hands-on time delivers quantitative protein binding kinetic data straight from expression mixtures, bypassing both binder purification and concentration determination.
