Formaldehyde Induced Leukemia-specific Chromosomal Aneuploidy
Although epidemiological evidence and toxicological studies support the association of formaldehyde and leukemia, to date, few studies have directly examined leukemia-specific changes on the causal pathways and have specifically targeted the hematopoietic stem/progenitor cells (HSCs/HPCs) of the exposed people. To address these critical issues, a molecular biomarker study was specially designed and conducted to explore the ability of formaldehyde to induce leukemia-specific chromosomal aneuploidy in the circulating HSCs/HPCs of exposed workers through two approaches: an initial approach and an expanded approach. The initial study primarily examined the formaldehyde-induced aneuploidy of chromosomes 7 and 8, frequently observed in leukemia patients. The expanded study employed a distinct chromosome-wide aneuploidy study (CWAS) approach to simultaneously evaluate aneuploidy of all 24 human chromosomes in the cultured circulating myeloid HSCs/HPCs of exposed but otherwise healthy Chinese workers. In this chapter, the findings and implications of both of the studies are detailed.
