Applications of Self-assembled Metallomacrocycles I: Biological Applications

Biomedical applications of self-assembled metallomacrocycles, especially for drug delivery and as anticancer agents, have grown exponentially in the last decade. The search for new chemical entities to overcome the limitations of commercial anticancer drugs, such as cisplatin, and the promising anticancer potentials of ruthenium-based small molecules, have prompted studies on transition-metal-directed self-assembled macrocycles with focus on their biomedical applications. As a result, a growing number of two- and three-dimensional metallomacrocycles that show promise for biological applications have been self-assembled. This chapter focuses on biological studies undertaken on the in vivo and in vitro characteristics of metallomacrocycles as regards their potential usage as anticancer drugs, DNA and protein-binding agents, or drug delivery agents. Future perspectives and limitations of these metallomacrocycles for biological applications are also briefly discussed.
