One-Sequence Mechanochemical Synthesis-Deracemisation of a Levetiracetam Intermediate

Abstract

Starting from a racemic building block, we introduce a one-pot mechanochemical synthesis of enantioenriched 2-(benzylideneamino)butanamide, a key intermediate to the enantipure drug levetiracetam. Optimisation leads to an ee of up to 94% ee with an overall 80% isolated yield.
