Unlocking a Modular Gd-DOTBA Platform via Tetramaleimide Click Chemistry for High-Performance MRI

Abstract

A tetramaleimide-functionalized Gd-DOTBA scaffold (Gd-DOTBA-Mal) is developed as a modular platform for high-performance MRI probes. The maleimide groups enable site-specific bioconjugation. A cysteine conjugate exhibits high relaxivity and stability, enabling sensitive kidney dysfunction assessment and tumor imaging at half the clinical dose. This versatile platform facilitates advanced molecular MRI development.
