Self-Propagating Amplification with a ss-Polyadenine Probe Enables Label-Free Detection of Terminal Deoxynucleotidyl Transferase

Abstract

We developed a label-free, self-propagating amplification strategy for ultrasensitive Terminal deoxynucleotidyl transferase (TdT) detection. The system integrates TdT-triggered poly-T extension, poly-A–primed synthesis, nicking-mediated recycling, and SYBR Green I–based fluorescence output, achieving robust signal amplification and assay performance.
