Chiral pool-engineered homochiral covalent organic frameworks for catalytic asymmetric synthesis of drug intermediate

Abstract

A chiral-pool strategy was employed to construct nanofiber-like cysteine-derived thiazoline COFs featuring a two-dimensional eclipsed (AA) stacking arrangement. The Cu(II)@(R)-THZTp achieved 95% yield and 93% ee in asymmetric Henry reactions. Integration with chitosan aerogel enabled gram-scale synthesis of R-salmeterol intermediate (92% yield, 93% ee) with excellent recyclability.
