Titania coated 2D gold nanoplates as nanoagents for synergistic photothermal/sonodynamic therapy in the second near-infrared window

Abstract

The development of efficient nanomedicine to improve cancer therapeutic effect is highly attractive. In this work, we firstly report titania coated Au nanoplate (Au NPL@TiO2) heterostructures, which play dual roles as nanoagents for synergistic photothermal/sonodynamic therapy in second near-infrared (NIR) window. On the one hand, since controlled TiO2 shell endows Au NPL@TiO2 nanostructures red-shifted to NIR II region, as-prepared Au NPL@TiO2 nanostructures posse high photothermal conversion efficiency of 42.05% irradiated by 1064 nm laserand are anticipated to be very competent candidates as photothermal agents. On the other hand, Au nanoplates (Au NPLs), as electron traps, vastly improve reactive oxygen species (ROS) generation of Au NPL@TiO2 nanostructures in contrast with pure shell TiO2 nanoparticles upon ultrasound (US) activation through sonodynamic process. Moreover, the toxicity and therapeutic effect of Au NPL@TiO2 nanostructures are relative systemically evaluated in vitro. Au NPL@TiO2 nanostructures generate a large amount intracellular ROS and exhibit laser power density-dependent toxicity, eventually inducing cancer cell apoptosis. Furthermore, synergistic therapeutic effect with only 20.3% cell viability upon both photothermal and sonodynamic activation is achieved at low concentrations of Au NPL@TiO2 nanostructures. The mice experiments also demonstrate the superiority of the combination of PTT and SDT with the totally tumor elimination. This work provides a way of two-dimensional (2D) gold nanoplate core/TiO2 shell nanostructures as novel nanoagents for advanced multifunctional cancer therapies in second NIR window.
