Ultrasensitive detection of H. pylori in human beings feces based on Immunomagnetic Beads capture and fluorescent quantum dots

Abstract

Given that Helicobacter pylori (H. pylori) was generally infected in early childhood and will exit in their whole lives without antibiotic treatment, it is quite convenient to use non-invasive methods to detect H. pylori for children. One of Stool antigen tests is an effective non-invasive method. This study presents a novel fecal test to detect H. pylori based on Immunomagnetic Beads (IMBs) with monoclonal antibodies sensitively recognizing and capturing the H. pylori, coupled with polyclonal antibodies -conjugating quantum dots probe, and ultrasensitive detection was achieved by fluorescence spectrometer. The detection method took 120 min with a limit of detection of 10^2 CFU/mL, and the linear ranged from 10 to 10^6 CFU/mL (R^2=0.9962). Most importantly, it can be effectively applied to real samples. This study provides a novel method for the detection of non-invasive fecal antigen H. pylori.
