Undefeatable relation between protein and osmolyte: A choice of nature


Many studies aimed at advancing our understanding of the mechanisms of protein stabilization that lead to survival of organisms under harsh environmental conditions, focus on deciphering the role of osmolytes as additives in stabilization of protein. The interaction of osmolyte with the protein is a matter of debate from a long time, however, still a clear unifying statement cannot be provided regarding the behavior of osmolyte. A literature survey reveals that there exist tremendously a large number of scholarly articles as well as elegant reviews which may lead to resolution of some problems of systems comprising of osmolyte and protein. Additionally, there is no doubt that vast literature survey indicates stabilizing behaviors of osmolytes towards protein stability. However, a number of research articles are available in open literature which emphasize on the destabilizing effects of osmolytes on the protein stability and function. Therefore, a complete acquaintance of each osmolyte with respect to each protein is one of the most challenging tasks in the development of protein formulation and may be still craving to administer protein formulations through injection only. The lack of comprehensive evaluation of a broad range of these osmolytes in protein systems was really stimulating interest for the present perspective in this research field. To the best of our knowledge, this perspective delineates the most recent successful advances in the open literature and also on the basis of our research experience that will provide new strategies for current researchers in the field of protein stabilization.
