Synthesis of carbocyclic NAD+ containing a methylenebisphosphonate linkage for the investigation of ADP-ribosyl cyclase

Abstract

A stereospecific synthesis of carbocyclic NAD+ incorporating a methylenebisphosphonatelinkage in place of the natural pyrophosphate gives an analogue 4 of the natural coenzyme NAD+ 3a designed to act as an inhibitor of ADP-ribosyl cyclase and to resist non-specific phosphatase degradation.
