Self-propagating amplification with a ss-polyadenine probe enables label-free detection of terminal deoxynucleotidyl transferase
Abstract
We developed a label-free, self-propagating amplification strategy for ultrasensitive terminal deoxynucleotidyl transferase (TdT) detection. The system integrates TdT-triggered poly-T extension, poly-A-primed synthesis, nicking-mediated recycling, and SYBR Green I-based fluorescence output, achieving robust signal amplification and assay performance.
