Amplified visual detection of microRNA-378 through a T4 DNA ligase-mediated circular template specific to target and target-triggering rolling circle amplification

Abstract

MicroRNA-378 (miRNA-378) is widely regarded as a novel noninvasive serum biomarker for early detection of gastric cancer. In the current study, a T4 DNA ligase-mediated circular template specific to target and a target-triggering rolling circle amplification (RCA) strategy was proposed for gastric cancer biomarker miRNA-378 visual detection. After multiple signal amplification through RCA and DNAzyme, a simple method for colorimetric detection of miRNA-378 was first developed with an ultralow detection limit of 80 fM per reaction by the naked eye, and synergetic signal cascade amplification enables the assay to have extraordinarily high sensitivity and the potential for clinical diagnosis.
