Seven new triterpenoid saponins from Astragalus membranaceus var. mongholicus and the inhibition of high-glucose induced SV40 MES 13 cells

Abstract

A phytochemical study of the ethanol extract of Astragalus membranaceus (Fisch) Bge. var. mongholicus (Bge) Hsiao resulted in the isolation of seven new triterpenoid saponins, astralanosaponins F–L, together with fifteen known compounds (8–22). Structure analysis of these extracted compounds was performed using 1D-NMR and 2D-NMR spectroscopy, and their data were compared with those of similar compounds previously reported in the literature. The absolute configuration of sugar was determined using GC-MS. The cell viability of SV40 MES 13 cells induced by high glucose was tested using the CCK-8 assay. Compound 19 showed obvious inhibitory effects of the high-glucose induced SV40 MES 13 cells with IC50 values of 12.2 ± 1.29 μM, while compounds 5 and 9 showed moderate inhibitory effects with IC50 less than 30 μM. Their possible structure–activity relationships were preliminarily summarized.
