NIR triggered release of DOX from sophorolipids coated mesoporous carbon nanoparticle with phase change 1-tetradecanol to treat MCF-7/ADR cell

Abstract

In order to prevent premature drug release from nanoparticles, it is vital to design and prepare a controlled and site-specific drug release system. We investigated a new controllable drug release mesoporous carbon nanoparticle (MCN) based nano-system that could take advantage of enhanced permeability and retention effect (EPR) to target tumor followed by the NIR induced heat triggered release of the chemotherapeutic drug to show anti-tumor effect. The pores of MCN with photo-thermal effects were filled with a chemotherapeutic agent doxorubicin (DOX) and a phase change material (PCM) 1-tetradecanol was used as gatekeeper to trap DOX inside the pores of mesoporous carbon nanoparticle as well as trigger the release of DOX inside the tumor cell using NIR irradiation. The surface of MCN was coated with natural Sophorolipid (SLPD) to obtain the resultant nanoparticles (DOX-PCM@MCN-SLPD) which have good biocompatibility, dispersibility and stability in aqueous solutions. The MCN based nano-system has the ability to load 24% DOX trapped with PCM before coating with SLPD. In comparison, it was observed that the release of DOX was temperature dependent above the melting point of PCM. Once the DOX-PCM@MCN-SLPD had been delivered into MCF-7/ADR cells, the release of DOX was triggered by MCN borne photo-thermal effects under NIR irradiation. The amount of DOX inside tumor was visualized by confocal laser scanning microscope, which shows higher amount of DOX at higher temperature as compared to normal body temperature, which further aids in the confirmation of minimization of pre-mature drug release at 37oC.
