β- Cyclodextrin based pH and thermo-responsive biopolymeric hydrogel as dual drugs carrier

Abstract

In the present study, chemically crosslinked, biocompatible and stimuli-responsive biopolymeric network gel has been developed to investigate its suitability as carrier for controlled release of dual drugs - metronidazole and ofloxacin, simultaneously. The crosslinked hydrogel [β-CD-cl-(PNIPAm-co-PMAc)] has been synthesized by grafting poly (N-isopropyl acrylamide) and poly (methacrylic acid) followed by crosslinking onto cyclic oligosaccharide β-cyclodextrin. β-CD-cl-(PNIPAm-co-PMAc) hydrogel has been characterized in details by several characterization techniques. The developed hydrogel reveals excellent pH and temperature responsive behaviour. Viscoelastic properties of the gel have been ascertained by measurement of rheological parameters. The network hydrogel displays non-cytotoxic nature towards MG-63 cell lines. The simultaneous in vitro release of metronidazole and ofloxacin from the gel endorses that it released both the colonic drugs at preferred pH (colonic pH 7.4) at a desired rate. The results manifest that β-CD-cl-(PNIPAm-co-PMAc) would be a promising entrant for targeted and sustained release of metronidazole and ofloxacin in vivo.
