An electrochemiluminescence sensor with dual signal amplification of Ru(bpy)32+ based on PtNPs and glucose dehydrogenase for diagnosis of gas gangrene
Abstract
Gas gangrene is often caused by Clostridium perfringens. Rapid detection of C. perfringens with point of care testing (POCT) can improve the therapeutic effect and save more patients' lives. Here, we report an electrochemiluminescence sensor based on a screen-printed electrode (SPE) and effective electrochemiluminescence (ECL) promotion of glucose dehydrogenase (GDH) for Ru(bpy)32+. In this strategy, platinum nanoparticles (PtNPs) could increase the amount of immobilized antibody and amplified the ECL signal. NADH produced by GDH was an effective co-reactant for Ru(bpy)32+. As a result, the sensitivity of the SPE sensor was enhanced by this dual signal amplification. The linear range of the SPE sensor assay is from 1 × 103 to 1 × 107 CFU mL−1 (colony forming units), with a lowest detection limit of 1 × 102 CFU mL−1. Results also suggested that it has excellent stability, reproducibility, and selectivity. It was suitable for the clinical diagnosis of gas gangrene in POCT.
