Light-Triggered Release of Drug Conjugates for Efficient Combination of Chemotherapy and Photodynamic Therapy

Abstract

Combination therapy is an emerging field in cancer treatment and have showed promises in providing synergistic efficacy outcome. Herein, we present a series of light-triggered porphyrin-based polymeric drug conjugates PSDTD-m for combined chemo-photodynamic therapy of cancer. PSDTD-m are covalently conjugated with both chemotherapy drug and photosensitizer. The chemodrug doxorubicin (DOX) was conjugated to PSDTD-m through ROS responsive linker that would be cleaved by ROS generated from adjacent photosensitizer under light irradiation. PSDTD-m self assemble into sub-100 nm micelles and showed good dark stability. Under light irradiation, on-demand DOX release and micelle disassembly was characterized. The ROS-cleavable controlled release of drug combined photodynamic therapy showed a higher anticancer efficacy, proving the effectiveness of this light triggered smart nanocarrier for enhancing therapy efficacy.
