The development of cell-initiated degradable hydrogel basing on methacrylated alginate applicable to multiple microfabrication technologies

Abstract

Biomimetic multicellular complex tissues can be achieved through 3D microfabrication approaches, while the lack of active materials that both meet the needs of molding and maintaining cell function is the bottleneck of its application. The development of 3D cell culture platform, suitable for multiple cell types and various microfabrication technologies, would bring a dawn to biomimetic construction of multicellular complex tissues. In this study, an MMP sensitive photocrosslinkable hydrogel system basing on methacrylated alginate, suitable for multiple cell survival and can be applied to a variety of micro-manufacturing technology, was designed for 3D microfeatures construction to meet the demand of both molding and biomimetic function requirement. Basing on the characteristics of photo-crosslinking and ion-crosslinking, the gel system was combined with a variety of microfabrication techniques to construct the biomimetic microstructure with spatial distribution of multiple cells. The shape diversity and cell universality of the assemblies, as well as cell activity maintenance, all show that the functional MMP-sensitive gel system has the potential to be used as a cell culture platform for tissue microstructure construction.
