Photo-Controlled Cell-Specific Metabolic Labeling of RNA

Abstract

Herein we present a chemical method for photo-controlled metabolic labeling of RNA. Our technique relies on photo-“caging” a metabolic intermediate. Once un-“caged” with long-wavelength UV light, the nucleoside analog can smoothly be incoroprated into cellular RNA. We anticipate this method will find wide-spread use in the field as it permits much-needed kinetic control over cell-specific RNA labeling experiments critical for understanding RNA expression programs.
